ABSTRACT

OBJECTIVE@#The study explores the effects of electroacupuncture (EA) at the governing vessel (GV) on proteomic changes in the hippocampus of rats with cognitive impairment.@*METHODS@#Healthy male rats were randomly divided into 3 groups: sham, model and EA. Cognitive impairment was induced by left middle cerebral artery occlusion in the model and EA groups. Rats in the EA group were treated with EA at Shenting (GV24) and Baihui (GV20) for 7 d. Neurological deficit was scored using the Longa scale, the learning and memory ability was detected using the Morris water maze (MWM) test, and the proteomic profiling in the hippocampus was analyzed using protein-labeling technology based on the isobaric tag for relative and absolute quantitation (iTRAQ). The Western blot (WB) analysis was used to detect the proteins and validate the results of iTRAQ.@*RESULTS@#Compared with the model group, the neurological deficit score was significantly reduced, and the escape latency in the MWM test was significantly shortened, while the number of platform crossings increased in the EA group. A total of 2872 proteins were identified by iTRAQ. Differentially expressed proteins (DEPs) were identified between different groups: 92 proteins were upregulated and 103 were downregulated in the model group compared with the sham group, while 142 proteins were upregulated and 126 were downregulated in the EA group compared with the model group. Most of the DEPs were involved in oxidative phosphorylation, glycolipid metabolism and synaptic transmission. Furthermore, we also verified 4 DEPs using WB technology. Although the WB results were not exactly the same as the iTRAQ results, the expression trends of the DEPs were consistent. The upregulation of heat-shock protein β1 (Hspb1) was the highest in the EA group compared to the model group.@*CONCLUSION@#EA can effect proteomic changes in the hippocampus of rats with cognitive impairment. Hspb1 may be involved in the molecular mechanism by which acupuncture improves cognitive impairment.

ABSTRACT

To explore the clinical distribution and drug resistance characteristics of carbapenem-resistant Klebsiella pneumoniae (CRKP), in order to provide reference for the prevention and treatment of CRKP infection. Retrospective analysis was performed on 510 clinical isolates of CRKP from January 2017 to December 2021, and strain identification and drug sensitivity tests were conducted by MALDI-TOF mass spectrometer and VITEK-2 Compact microbial drug sensitivity analyzer. The carbapenemase phenotype of CRKP strain was detected by carbapenemase inhibitor enhancement test. The CRKP strain was further categorized by immunochromogenic method and polymerase chain reaction (PCR) was used for gene detection. The results showed that 302 strains (59.2%) were derived from sputum, 127 strains (24.9%) from urine and 47 strains (9.2%) from blood. 231 (45.3%) were mainly distributed in intensive care, followed by 108 (21.2%) in respiratory medicine and 79 (15.5%) in neurosurgery. Drug susceptibility test result shows that the resistant rate of tigecycline increased from 1.0% in 2017 to 10.1% in 2021, the difference was statistically significant (χ2=14.444,P<0.05). The results of carbapenemase inhibitor enhancement test showed that 461 carbapenemase strains (90.4%) of 510 CRKP strains, including 450 serinase strains (88.2%), 9 metalloenzyme strains (1.8%), and 2 strains (0.4%) produced both serine and metalloenzyme. 49 strains (9.6%) did not produce enzymes. Further typing by immunochromogenic assay showed that 461 CRKP strains were KPC 450 (97.6%) and IMP 2 (0.4%). 7 NDM (1.5%); 2 strains of KPC+NDM (0.4%); PCR results were as follows: 450 strains of blaKPC (97.6%), 2 strains of blaIMP (0.4%), 7 strains of blaNDM (1.5%), and 2 strains of blaKPC+NDM (0.4%). In conclusion, CRKP strains mainly originated from sputum specimens and distributed in intensive care department, and the drug resistance characteristics were mainly KPC type in carbapenemase production. Clinical microbiology laboratory should strengthen the monitoring of CRKP strains, so as to provide reference for preventing CRKP infection and reducing the production of bacterial drug resistance.

ABSTRACT

ObjectiveTo explore the effect of Qishengwan on ileal flora during its treatment of Alzheimer's disease (AD) under the guidance of the theory of "interior-exterior relationship between heart and small intestine". MethodThe AD model was established by bilateral intraventricular injection of β-amyloid 1-42 (Aβ1-42). The rats were then randomly divided into the blank group, sham-operated group, model group, low-, medium-, and high-dose (5.6, 11.2,22.4 g·kg-1·d-1) Qishengwan groups, and donepezil (0.46 mg·kg-1·d-1) group. After medication for 28 successive days, the spatial memory ability of rats was observed in water maze test, and the levels of Aβ1-42, nuclear transcription factor-κB (NF-κB), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in the hippocampus were analyzed by enzyme-linked immunosorbent assay (ELISA). Additionally, the contents of the ileum were collected and subjected to 16SrRNA-sequencing analysis for figuring out the changes in ileal flora. ResultCompared with the blank group and sham-operated group, the model group exhibited significantly reduced stay time in the target quadrant and number of target quadrant and platform crossings (P<0.05, P<0.01) and elevated Aβ1-42 content in the hippocampus (P<0.01) and central inflammatory factors NF-κB, TNF-α, and IL-6 (P<0.05, P<0.01). Compared with the model group, Qishengwan at each dose significantly alleviated the impaired spatial memory function (P<0.05, P<0.01), improved the deposition of Aβ1-42 in the hippocampus of rats (P<0.05, P<0.01), and reduced the expression of central nervous system inflammatory factors (P<0.05, P<0.01), thus exerting a good therapeutic effect on AD rats. The 16SrRNA-sequencing analysis results showed that the structure of the ileal flora in the model group was significantly separated from those in the blank group and sham-operated group. The abundance of Lachnospiraceae NK4A136 group was significantly increased (P<0.01), while that of Escherichia-Shigella was reduced (P<0.05, P<0.01). Qishengwan at each dose significantly changed the ileal flora structure and regulated the relative abundance of Lachnospiraceae NK4A136 group, Escherichia-Shigella, and Ruminococcaceae. ConclusionQishengwan has a positive therapeutic effect on AD. It can significantly enhance the memory and cognitive abilities in AD rats, which may be related to its regulation of the structure of rat ileal flora and the relative abundance of Lachnospiraceae NK4A136 group, Escherichia-Shigella, and Ruminococcaceae, the attenuation of the central neuroinflammatory response, and the reduction of central Aβ1-42 deposition.

ABSTRACT

@#Abstract: Objective To analyze the survival status of HIV/AIDS patients aged above 50 years receiving antiviral therapy (ART) in Shanxi Province from 2011 to 2019, and to provide evidence for further improvement of antiviral therapy. Methods Basic information and follow-up information of HIV/AIDS patients aged above 50 years who first received HIV/AIDS antiviral therapy in Shanxi Province from 2011 to 2019 were collected. Excel database was established and SPSS23.0 software was used for analysis. Retrospective cohort study was conducted. Cox proportional risk regression model was used to analyze the factors influencing survival time. Results A total of 1 183 subjects were included, of which 172 died, including 84(48.84%) from other causes, 74(43.02%) AIDS-related death and 14 (8.14%) from accidents, suicides and undetermined deaths. Setting AIDS-related deaths as an outcome event, life table analysis showed that the cumulative survival rates at 1, 3, 5, 7 and 9 years after receiving ART were 96.61%, 93.59%, 90.35%, 87.57% and 83.44%, respectively. Multivariate Cox proportional risk model analysis showed that the risk of death in patients aged 60-<70 years group and over 70 age group was 2.53 times (95%CI: 1.51-4.23) and 3.59 times (95%CI: 1.74-7.40) for patients aged the 50-<60 group , respectively. The risk of death in patients with baseline CD4+T lymphocyte (CD4) counts of ≥200/mm3, 50-<200 /mm3 was 0.22 times (95%CI: 0.12-0.41) and 0.37 times (95%CI: 0.21-0.67) for patients with CD4+T lymphocyte counts of <50/mm3. The risk of death in patients with opportunistic infections at baseline was 1.99 times (95%CI: 1.16-3.39) for patients without baseline opportunistic infections. Conclusions The survival rate of HIV/AIDS patients aged above 50 who received antiviral therapy (ART) in Shanxi Province from 2011 to 2019 was relatively high. To further improve the quality of antiviral treatment in our province, the strategy of "early detection and early treatment" should be continued and improved in the future, and information collection of specific causes of non-AIDS-related deaths among this population should be further strengthened.

ABSTRACT

ObjectiveTo evaluate the dosage effect of measles, mumps and rubella combined attenuated live vaccine (MMR) vaccination on seroprevalence of mumps. MethodsA cross-sectional study was conducted among people in Changning District of Shanghai aged 1 month to 19 years old (n=1 816) in Mar.-Sep. 2017. Blood samples were analyzed for mumps antibodies using enzyme-linked immunosorbent immunoglobulin G (IgG) assays. ResultsMumps antibody seropositivity was 94.59% in 2 years old children and maintained at 98.18%-100.00% from 4 to 9 years old. The seropositivity began to decrease since 10 years, and it was 88.33% (95%CI: 81.20%-93.47%) at age of 12 years. In 12-19 years age group, individuals with 3 doses of mumps-containing vaccines had the highest seropositivity (93.88%) and individuals with 1 or 0 doses had the lowest seropositivity (68.75%). ConclusionTwo-dose MMR immunization in Shanghai induces a sharp increase in mumps antibody levels in the corresponding age groups. The antibody levels decline gradually with time since the second dose. Vaccine dosage is positively associated with mumps IgG seropositivity and geometric mean concentrations (GMC) in 12-19 years old.

ABSTRACT

OBJECTIVE@#To establish a method for the production of digital individual tooth tray based on three-dimensional (3D) scan, computer-aided design (CAD) and 3D printing, and to evaluate the effect of impression taking of full-arch crown abutments by digital individual tooth tray technique and conventional method through in vitro study.@*METHODS@#The full crown preparation was performed on all the fourteen resin teeth in a standard model of mandibular dentition. The surface data of prepared abutments was collected by 3D scanning. A new project was created in a dental CAD software including all the fourteen teeth in the mandibular dentition. The design modules of anatomy crown and coping were selected for each tooth. The dentition was divided for three sections: right posterior teeth, anterior teeth, and left posterior teeth areas. The connector design was added between the abutments within the same section. The scanned data of the abutments were imported. The occlusal plane and insertion path were determined. The position of margin line, as well as the shape of anatomy crown and connector as the main body of the individual tooth tray were designed for each abutment. The shape of coping was generated as the space for holding the impression material. The finalized data of the main body was imported into Geomagic software. The retentive attachment was added at the external surface and the tissue stop was formed at the internal surface. The completed individual tooth tray was manufactured by 3D printing with resin material. The data of full-arch crown abutments were modified and printed. The conventional dentition trays A and B, as well as digital individual tooth tray were designed and printed for four copies each. The polyether impressions of the full-arch abutments were made by conventional one-step method using dentition tray A, and by sectional-impression technique using digital individual tooth tray and dentition tray B for four times each. The time spent for each impression taking and the numbers of defects at the shoulder and axial/occlusal surface in each impression were recorded. The impression quality of each abutment was evaluated. The overall quality distribution and the pass rate of abutments between the two methods were analyzed.@*RESULTS@#The impressions made by conventional method had more defects at shoulder than those made by digital individual tooth tray technique. No difference of the number of defects at axial/occlusal surface between the two methods was observed. The digital individual tooth tray technique for the full-arch abutment impression exhibited higher pass rate of abutments and better quality of impression, compared with conventional methods.@*CONCLUSION@#A new method for the production of digital individual tooth tray based on digital scanning, CAD and 3D printing was established. Compared with conventional method, using digital individual tooth tray technique for impression taking of full-arch abutments can achieve better effect.

ABSTRACT

Pancreatic cancer is one of the cancers with poor prognosis at present, which has high resistance to various anti-tumor drugs as a result of the interaction among pancreatic cancer cells, cancer stem cells, and the tumor microenvironment. P2X7 receptors are extracellular adenosine triphosphate(ATP)-gated nonselective cation channels, which have many biological functions including being involved in cell signal transduction and cytokine secretion, mediate cell survival and growth. Studies have shown that P2X7 receptor is highly expressed in pancreatic cancer and promotes the proliferation, migration and invasion of pancreatic cancer cells by supporting proliferation of pancreatic stellate cells and regulating the expressed of the MMP2/MMP9 protein. The paper reviews the recent research advances of P2X7 receptor in pancreatic cancer.

ABSTRACT

OBJECTIVE@#To compare the orofacial pain sensitivity with operant test and mechanical hyperalgesia with von Frey filaments of two orofacial pain models (EOI: experimental occlusal interference; pIONX: partial infraorbital nerve transection). To investigate the operant and evoked characteristics of EOI-rats.@*METHODS@#The orofacial operant behaviors were tested by Ugo Basile Orofacial Stimulation Test System. The mechanical thresholds of vibrissal pads were tested by von Frey filaments. Male Sprague-Dawley rats were randomly divided into eight groups: von Frey group: sham-EOI, EOI, sham-pIONX, pIONX (sham: sham-operated group); operant test group: sham-EOI, EOI, sham-pIONX, pIONX (sham: sham-operated group). The mechanical thresholds and orofacial operant behaviors were tested on pre-operation and post-operation days l, 3, 7, 10, 14 and 21.@*RESULTS@#In pIONX of von Frey group, the mechanical withdrawal threshold decreased from days 1 to 21 (P<0.05), peaking from days 7 to 10, and lasted until the end of the experiment. There was no significant difference between the bilateral sides. In pIONX of operant test group, the total contact time decreased from days 10 to 21 (P<0.05), peaking from days 10 to 14, and lasted until the end of the experiment. In EOI of von Frey group, the mechanical withdrawal threshold decreased from days 3 to 21 (P<0.05), peaking on day 7, and lasted until the end of the experiment. There was no significant difference between the bilateral sides. In EOI of operant test group, the total contact time decreased from days 1 to 21 (P<0.05), peaking from days 7 to 10, and lasting until the end of experiment.@*CONCLUSION@#Orofacial operant test is a stable method to evaluate orofacial pain behaviors, which could discriminate the feature of neuropathic and EOI orofacial pain. In these two animal models, both of the operant behaviors and the mechanical hyperalgesia exhibited different time courses. Orofacial operant test provides a novel method for evaluating the orofacial pain sensitivity and studying the orofacial pain mechanism thoroughly.

ABSTRACT

As the P2X receptor family has become a research hotspot in recent years, the study on P2X7 receptor has also received extensive attention. P2X7 receptor (P2X7R) is a dual-transmembrane cation-channel receptor, which is gated in vivo by adenosine triphosphate (ATP) and can be activated. Activated P2X7 receptor can produce such effects as cationic channel opening, signal pathway activation, inflammatory mediator release and cell apoptosis. It plays an important role in the development of various tumors and inflammatory diseases. Studies have shown that the abnormal expression of P2X7 receptor affects the occurrence and development of breast cancer by activating a series of signaling pathways, which is expected to become a new target for designing anti-breast cancer drugs. Other studies have shown that multiple microRNAs can promote the occurrence and development of breast cancer by regulating the expression of P2X7 receptor gene. In this review, we reviewed the recent research advances of P2X7 receptor and the relationship between breast cancer and its abnormal expression.

ABSTRACT

Penicillin-binding proteins (PBPs) are the target of β-lactam antibiotics (the major treatment for Streptococcus pneumoniae infections), and mutations in PBPs are considered as a primary mechanism for the development of β-lactam resistance in S. pneumoniae. This study was conducted to investigate the mutations in the PBPs of clinical S. pneumoniae isolates in Hangzhou, China, in correlation with β-lactam resistance. Results showed that 19F was the predominant serotype (7/27) and 14 of the S. pneumoniae isolates were resistant to both penicillin G and cephalosporin. Genotyping results suggested that β-lactam-resistant isolates primarily exhibited single-site mutations in both the STMK and SRNVP motifs of pbp1a in combination with double-site mutations in the STMK motif of pbp2x, which might be the primary mechanisms underlying the β-lactam resistance of the isolates in this study.

ABSTRACT

Purpose To study the clinicopathologic features of ganglioglioma. Methods The clinicopathologic data of the cases pathologically diagnosed as ganglioglioma that underwent resection of epileptic focus were retrospectively analyzed. Results In the 19 cases studied, the mean onset age was 9.1 years, and the duration of disease was 9.3 years. MRI images showed abnormal signals. The majority of the site was temporal lobe (14/19, 73.7%). The tumors showed heterogeneity and often accompanied by focal cortical dysplasias (13/19, 68.4%). Immunohistochemical staining showed CD34 positive in 18 cases, Nestin positive in 16 cases, and BRAF-V600E positive in 6 case. The positive expression rate of CD34 and Nestin did not have significant differences. Conclusion The diagnosis of ganglioglioma relies on pathological observations combined with clinical features and neuroradiological examinations. Differential diagnosis should be done from other tumors or cortical dysplasia. Immunohistochemical staining of CD34 and Nestin can help diagnosis.

Objective To carry out a quantitative estimate that related to the effects of shortterm exposure to PM2.5 on all-cause mortality and emergency visits in China by using the systematic review and Meta-analysis.Methods We selected all the studies published before March 2018 from China National Knowledge Infrastructure,Wanfang database,PubMed and EMBASE and data on relative risk (RR),excess risk (ER) and their 95%CIs:appeared in these papers were extracted.According to the differences in the size or direction (heterogeneity) of the results,we computed summary estimates of the effect values using a random-effect or fixed effect model. We also conducted the subgroup analysis and Meta-analysis to have assessed the selected studies for the evidence of study bias. Results A total of 33 original studies, indexed in databases, were identified.Among those studies,39 sets of data on mortality and 4 sets of data on emergency were valid to show that within the daily concentration range from 47.7 to 176.7 pg/m3,for 10 μg/m3 increases in PM2.5 concentrations,it would increase the daily numbers of deaths by 0.49% (95%CI:0.39%-0.59%) and 0.30% (95%CI:0.10%-0.51%) for all-cause deaths and all-cause emergency-room visits, respectively. For subgroup analysis, the combined effect of PM2.5 in causing short-term all-cause deaths in the northern areas (ER=0.42%,95%CI:0.30%-0.54%) seemed lower than that in the southern areas (ER=0.63%,95%CI:0.44%-0.82%).The combined effect of PM2.5 concentration below 75 μg/m3 (ER=0.50%,95%CI:0.37%-0.62%) was higher than that of PM25 concentration ≥75 μ g/m3 (ER=0.39%,95% CI:0.26%-0.52%).Conclusion Within the concentration range from 47.7 to 176.7 μg/m3,short-term exposure to current level of PM2.5 might increase both the all-cause daily mortality and daily emergency visits in China.

ABSTRACT

Rheumatoid arthritis (RA) is a chronic inflammatory joint dysfunction by inflammation driven autoimmune disease, with joint swelling, pain and stiffness, resulting in severe disability, while increasing the risk of suffering from extra ar-ticular disease.Rehabilitative therapy,as an auxiliary intervention beyond drug therapy, plays an important role in RA treatment, which includes physical therapy, kinesitherapy and occupational therapy. Physical therapies are effective on improving the symptom of RA and alleviating inflammation. Kinesitherapy could reduce inflammatory reaction, im-prove the joint function, reduce the risk of cardiovascular disease, and improve the mental health.Occupational therapy may help them adapt the living and working status, and improve the quality of life.

ABSTRACT

Objective To analyze the surveillance results of soil-borne nematodiasis in Hefei City from 2012 to 2015,so as to provide the evidence formulating the Hefei"Thirteen-five"parasitic disease prevention strategy. Methods The population in monitoring points in Changfeng County,Lujiang City and Chaohu County were investigated for Ascaris lumbricoides,Trichuris trichiura and hookworm infections with Kato-Katz technique every year from 2012 to 2015,and the children aged less than 6 years were checked for Enterobius vermicularis infection by using the cellophane tape method. Results A total of 12 496 people were investigated from 2012 to 2015,the number of infections was 84,and among them, the number of hookworm infection was the most(70 cases)and the number of A.lumbricoides was the least(3 cases). The infection rate in 2014 was higher than that in other years, and there was a statistically significant difference(P<0.008).There was a statistically significant difference among all age groups over 6 years of age in the infection rates(Χ2=27.229,P<0.05),and the infection rate of the age group over 66 years was higher than that of the other age groups. The infection rate in the female was higher than that in the male(Χ2=5.378, P<0.05).The infection rate in the monitoring point in Changfeng County was higher than that in the other monitoring points (P<0.017).Conclusions From 2012 to 2015,the infection rate of soil-borne nematode infections is lower than 5% in Hefei City. The number of hookworm infection is the most. Therefore, during the〝Thirteen five〝period, the key point of prevention of parasitic infections is hookworm infection.

ABSTRACT

<p><b>OBJECTIVE</b>To assess the clinical effects of artificial radial head prosthesis replacement for the treatment of comminuted fracture of the radial head.</p><p><b>METHODS</b>From June 2011 to June 2015, 25 patients with radial head comminuted fracture were treated with artificial radial head replacement, including 10 males and 15 females, ranging in age form 24 to 61 years old(mean, 40 years old). The functional recovery of the patients suffering from limb and elbow in different periods, the activity degree of the elbow joint and the function of the elbow in the latest follow-up were compared.</p><p><b>RESULTS</b>All the patients were followed up, and the duration ranged from 12 to 48 months, averaged 26 months. There were no complications such as infection, elbow instability, subluxation of the distal radioulnar joint, and myositis ossificans. The VAS, Broberg and Morrey elbow function score were improved 6, 9 months after operation compared with that 3 months after operation(<0.05). There were significant differences in elbow flexion and extension, rotation activity between injured side and healthy side 3, 6, 9 months after operation(<0.01), but no significant differences between injured side and healthy side at the latest follow-up(>0.05). At the latest follow-up, according to Broberg and Morrey elbow function evaluation criteria, 16 cases got an excellent result, 7 good and 2 poor.</p><p><b>CONCLUSIONS</b>It can maximize the recovery of elbow joint stability and quicken early functional exercise, prevent and reduce the occurrence of complications by using the artificial radial head replacement therapy to repair comminuted fracture of the radial head. The short-term curative effect is satisfactory, but the long-term effect needs further observation.</p>
